Fortify Alteration potions do one thing: any Alteration spell you cast while the potion is active lasts longer than normal. The boost runs for 60 seconds and ranges from a quarter extra duration on the weakest bottle to double duration on the elixir. Mages who lean on protective Alteration magic get the most out of them, since a longer-lasting ward or flesh spell means fewer recasts in the middle of a fight.

Where to find them
These are ordinary leveled potions. You can buy them from merchants who stock potions, and they turn up as loot in the world and in the inventories of other characters. Because they are leveled, the stronger tiers become more common as the Dragonborn gains levels. Every bottle weighs 0.5.
Brewing your own
With the Alchemy skill you can make these yourself. Only three ingredients carry the Fortify Alteration effect: Grass Pod, River Betty and Spriggan Sap. Any two of them combined will produce the potion.
Mixing at an alchemy lab follows the usual rule: two or more ingredients must share an effect before anything useful comes out. Eating an ingredient reveals one of its properties (more with the Experimenter perk), and once an effect is known you can pick it straight from the lab menu instead of guessing.
Tips
Drink the potion first and cast afterwards: the bonus applies to spells cast during its 60 seconds, so timing matters more than with a simple restore potion. If you also want to cast more often, pair it with Fortify Magicka Potions.
